Capacidade antioxidante in vitro e efeito hepatoprotetor in vivo do extrato de folhas de Allophylus edulis
| dc.contributor.advisor | Allophylus edulis is used in folk medicine primarily for liver conditions such as hepatitis, liver cancer and liver cirrhosis. In vitro hepatoprotective activity was previously demonstrated. The aim of this work was to evaluate the antioxidant capacity and hepatoprotective effect of the ethanolic extract of A. edulis in mice. This was done by first determining the acute toxicity of the extract, evaluating the general behavior, and subsequently verifying the effect on paracetamol-induced toxic hepatitis in male mice. Additionally, the phytochemical profile was performed, and the content of total phenols and its total antioxidant capacity were quantified through the 2,2, azinobis- (3-ethylbenzothiazoline)-6 sulfonic acid radical cation (ABTS) method. The extract of A. edulis leaves did not demonstrate adverse effects up to 2000 mg/kg, p.o. Anthraquinones, flavonoids, triterpenoids, and tannins were detected. A high content of total phenolic compounds (TPC) was related to a high antioxidant capacity. Regarding the results of the biological tests, A. edulis did not affect the general behavior of the mice, and all doses tested decreased glutamic-oxaloacetic transaminase (GOT) and glutamic-pyruvic transaminase (GPT) activity, the main liver enzyme markers of hepatocellular damage. It is concluded that A. edulis has hepatoprotective activity, which could be related to its antioxidant activity. | dc.description.abstract | Allophylus edulis é usado na medicina tradicional, principalmente em doenças do fígado, como hepatite, cancro e cirrose hepática. A atividade hepatoprotetora in vitro foi previamente demonstrada. O objetivo deste trabalho foi avaliar a capacidade antioxidante e o efeito hepatoprotetor do extrato etanólico de A. edulis em murganhos. Para tal, foi determinada a toxicidade aguda do extrato, avaliando o comportamento geral e, posteriormente, verificando o efeito sobre a hepatite tóxica induzida por paracetamol em murganhos macho. O perfil fitoquímico foi definido, e quantificado o teor de fenóis totais e a sua capacidade antioxidante total através do método do catião radical 2,2’-azino-bis (3-etilbenzotiazolin) 6-ácido sulfónico (ABTS). O extrato de folhas de A. edulis não demonstrou efeitos adversos até 2000 mg/kg, p.o. Foram detetados antraquinonas, flavonoides, triterpenoides e taninos. Um elevado teor de compostos fenólicos totais (TPC) foi relacionado com uma elevada capacidade antioxidante. Relativamente, aos resultados dos testes biológicos, A. edulis não afetou o comportamento geral em murganhos, e todas as doses testadas diminuíram a atividade da transaminase glutâmico-oxalacética (TGO) e da transaminase glutâmicopirúvica (TGP), os principais marcadores das enzimas hepáticas de dano hepatocelular. Conclui-se que A. edulis possui atividade hepatoprotetora, o que pode estar relacionado com a sua atividade antioxidante.
