O presente trabalho procura apresentar uma revisão bibliográfica sobre o tema geral das demências. Por haver várias doenças que se podem enquadrar no âmbito das demências, esta dissertação centra-se fundamentalmente na Doença de Alzheimer, devido à sua prevalência e ao aumento do número de casos a nível global.
O objetivo principal deste trabalho, foi fazer uma revisão bibliográfica relativamente ao mecanismo de ação da Doença de Alzheimer, as terapêuticas atuais e as potencialmente utilizadas, abordando as principais limitações do tratamento desta patologia no quadro atual do conhecimento. De modo a conseguir atingir estes objetivos procedeu-se a uma revisão bibliográfica da literatura científica, priveligiando o PubMed como motor de busca, cuja pesquisa se centrou na procura de artigos com 10 anos de publicação, e em investigação focada em humanos. Como pesquisa complementar recorreu-se também ao MeSH, ao Google Scholar e ao Web of Science. As palavras-chave utilizadas na pesquisa foram Alzheimer disease, alzheimer disease treatment, neurodegenerative diseases, physiopathology of alzheimer's disease, gene therapy in Alzheimer's disease e diagnosis of Alzheimer's disease.
No final deste trabalho, apresentam-se alguns tratamentos ou substâncias que podem servir para retardar o avanço da doença, e melhoramento das alterações comportamentais, embora nenhuma cura tenha sido encontrada.
The present work aims to present a review of the literature on the general theme of dementias. Because there are several diseases that can be classified as dementia, this dissertation focuses mainly on Alzheimer's disease, due to its prevalence and the increase in the number of cases globally. The main goal of this work was to make a review of the literature regarding the mechanism of action of Alzheimer's disease, current and potential therapies, while addressing the main limitations of the treatment of this pathology within the current state of the art. In order to achieve these goals, a bibliographic review of the scientific literature, based mainly on PubMed, was carried out, whose research focused on the search for articles with 10 years of publication, referring to humans. Search engines like MeSH, Google Scholar and Web of Science were also used. The keywords used in the research were Alzheimer's disease, alzheimer's disease treatment, neurodegenerative diseases, physiopathology of Alzheimer's disease, gene therapy in Alzheimer's disease and diagnosis of Alzheimer's disease. At the end of this paper, we present some treatments or substances that may serve to slow the progression of the disease, and improve behavioral changes, although no cure has been found.
