Histological characterisation of the NLrp3 inflammasome in the ovary of obese mice

A obesidade tem vindo a aumentar durante os últimos anos. Paralelamente, o número
de mulheres obesas com problemas reprodutivos tem vindo também a aumentar. Sabe-se que o
inflamassoma Nlrp3 está presente no estado inflamatório característico da obesidade e pela
produção e secreção de Il-1b, pelo que é plausível estabelecer uma relação entre a ativação do
inflamassoma e as alterações reprodutivas associadas à obesidade. Assim, este trabalho
caracteriza histologicamente a expressão do inflamassoma Nlrp3 no ovário de murganhos
obesos, tendo como controlo a expressão da Il-1b. Para tal, levou-se a cabo a técnica de
imunohistoquímica de forma a identificar as proteínas Nlrp3 e Il-1b em ovários dos seguintes
quatro diferentes grupos de murganhos: i) murganhos em que a obesidade foi induzida por dieta;
ii) murganhos ob/ob; iii) murganhos db/db; iv) murganhos injetados com leptina. No total,
foram observados 21 cortes de ovários de murganhos com o anticorpo do inflamassoma Nlrp3
e 22 cortes de ovários de murganhos com o anticorpo Il-1b. Com este trabalho, foi possível
concluir que o inflamassoma Nlrp3 está expresso nos oócitos de todos os folículos de cada fase
folicular, nas células da granulosa e no estroma de ovários obesos, o que comprova a associação
da inflamação com a obesidade.
Obesity has been increasing during the last years. Also, the number of obese women with reproductive disorders has been increasing. It is known that the Nlrp3 inflammasome is present in the inflammatory state characteristic of obesity and for the production and secretion of Il-1b. Therefore, it is plausible to establish the link between inflamassome activation and obesity related reproductive disorders. Thus, this study histologically characterises the expression of the Nlrp3 inflammasome in the ovary of obese mice, having as a control the expression of Il-1b. Therefore, it was performed immunohistochemistry staining of Nlrp3 and Il-1b proteins in ovaries collected from four diferente groups of mice as follows: i) diet-induced obesity mice; ii) ob/ob mice; iii) db/db mice; iv) leptin-injected mice. A total of 21 mice ovarian cuts with the Nlrp3 inflammasome antibody and 22 mice ovarian cuts with the Il-1b antibody were processed. With this study, it was possible to conclude that the Nlrp3 inflammasome is expressed in the oocytes of follicles of all stages of the follicle development, in the granulosa cells and in the stroma of obese ovaries, supporting the relation between inflammation and obesity.
